Way determination of the karyotype of sugar sorghum obtained in in vitro culture includes cell research at the stages of metaphase and anaphase， growing explants in in vitro， conducting prefixation effect on mitosis， use of 0.03% ortho-oxyquinoline to reduce chromosomes， selection of explants with a size of 2-3 mm， the use of acetoorsein 3%， washing with distilled water objects. At what use the growth points of sugar sorghum obtained in vitro， which carry out preliminary cold treatment - 12 hours， with: conditions and time staining - in a drop of dye on a glass slide - 5 minutes， cell division and chromosome reduction - 5 hours， maceration - 10 minutes.
