The aim of this study was to evaluate the microbial diversity of urban stream water and periphyton. The study took place along Third Creek in Knoxville, Tennessee, July 2021. DNA was extracted from twelve separate water samples that were paired with co-occuring periphyton samples using Qiagen kits, and the V4 region of the 16S rRNA gene (using 515F and 806RB primers for Bacteria and Archaea), and a section of the 18S rRNA gene (Euk1391 and EukBr primers for eukaryotes), were sequenced using the Illumina MiSeq platform.
