Age and skeletal maturity impacts the capacity for bone regeneration. Using a mouse model of large-scale rib resection, we demonstrated that skeletally immature mice regenerate the rib around 14 days post resection, whereas skeletally mature mice never fully regenerate the rib. Here, we performed bulk RNA sequencing analysis of rib calluses from skeletally immature and mature mice at baseline, 0, 2, 5, and 14 days post-resection to characterize the age-related changes in gene expression profiles during the early and late stages repair of bone repair. Rib calluses were dissected from the rib cages of young skeletally immature and old skeletally mature mice at different stages of rib regeneration (baseline, 0, 2, 5, and 14 days post-resection) and snap frozen. Frozen tissue was ground using a mortar and pestle kept in liquid nitrogen. Powdered frozen tissue was immediately transferred to a clear tube and total RNA was extracted using TriPure Isolation Reagent (Sigma-Aldrich) followed by PureLink™ RNA Mini Kit (Thermo Fisher Scientific).
