The skin is a vital barrier protecting the human body from the external environment. While cell type-specific transcriptional heterogeneity in human skin has been extensively documented, the underlying regulatory mechanisms remain largely unexplored. In this study, we employed high-throughput single-cell chromatin accessibility and transcriptome sequencing (HT-scCAT-seq), a technique for simultaneously profiling transcriptome and chromatin accessibility, to investigate these mechanisms. Using HT-scCAT-seq, we analyzed 10,065 single-cell profiles from human eyelid skin of four individuals. Our analysis revealed detailed molecular signatures defining 13 distinct cell types within eyelid skin and identified key transcription factors specifically enriched in each cell type. This comprehensive dataset serves as a valuable resource and provides a crucial foundation for future investigations into the molecular mechanisms governing human skin biology.
