The Andrechek lab has hypothesized a role for E2F5 in mammary gland development using scRNAseq data and predictive modeling. Whole body knockouts of E2F5 resulted in early lethality of mice due to hydrocephalus. To study the role of E2F5 specifically in the mammary gland and avoid early lethality, our lab has generated an MMTV-Cre inducible conditional knockout of E2F5 mouse model that ablates exons 2 and 3 of E2F5 in the epithelial cells of the mouse mammary gland. After a long latency period, highly metastatic mammary tumors developed with high penetrance. To identify the mechanisms of mammary tumor formation after loss of E2F5, our lab performed whole genome sequencing and bulk RNAseq on E2F5 knockout tumors.
