Abstract*: Mouse embryonic stem (ES) cells exhibit unlimited self-renewal capacity while maintaining pluripotency under appropriate culture condition. The study of the exit from pluripotency can be achieved by recessive screening with selection pressure of removing two small molecules (2i) and leukaemia inhibitory factory (LIF). To address this question we decided to adopt the method of genome-wide loss of function screening based on the recently discovered clustered regularly interspaced short palindromic repeats (CRISPR) â CRISPR-associated (Cas) systems. This study is to validate phenotype of known genes which are crucial for the dissolve of pluripotency circuit.
