EHEC Growth Curve RNA Sequencing

This sequencing was carried out to identify growth phase-dependent changes in gene expression in E. coli O157:H7. Â Total RNA was isolated at OD600 ~ 0.8 (tp6) and ~ 3.4 (tp10) from a replicate cultures of a derivative of E. coli O157:H7 EDL933 named ES669 constructed by Shakhnovich et al. (Molecular Microbiology, 2009). Ribosomal RNA was depleted and barcoded Illumina libraries were constructed from this RNA as described ( Giannoukos et al., Genome Biology 2012). Â These libraries were sequenced in a single lane of Illumina HiSeq 2000 multiplexed with several others to yield 36 base single reads.
