Nanorate sequencing libraries were created from 15 Arabidopsis thaliana plants and sequenced on the Illumina NovaSeq X series platform. 8 wild type sibling Col-0 plants and 1 Ler-0 plant were grown under standard long day conditions (untreated); 2 Col-0 siblings were grown under a UVB light which was on for 8 hours every day; and 4 Col-0 siblings were treated with multiple acute UVC irradiations (10x0.02J/cm2, 10x0.05J/cm2, 4x0.1J/cm2, 2x0.2J/cm2) over the course of growth. All plants were harvested at the opening of the first flower. For the Col-0 plants, flowers and flower buds were removed and the remaining shoot was flash frozen. For the Ler-0 plant, a single leaf was flash frozen. For the untreated Col-0 samples, the NanoSeq library was split into 3 replicates before PCR amplification. Sequencing data was used to identify somatic mutations in the Col-0 sample and germline substitutions in the Ler-0 sample.
