Rothia mucilaginosa intervention study: 16S rRNA gut microbiome and complete genome sequencing

This study investigates the effects of Rothia mucilaginosa intervention on the gut microbiome composition in mouse models using 16S ribosomal RNA gene amplicon sequencing. We analyzed gut microbiome samples from three groups: normal control (NC, n=8), model control (MC, n=8), and Rothia intervention group (RG, n=10), with a total of 26 samples. The constipation model was induced by daily gavage of loperamide (10-15 mg/kg) for 14 days, followed by intervention phase where the RG group received daily oral gavage of Rothia mucilaginosa suspension (2x10^8 CFU in 200 microL PBS) while NC and MC groups received PBS control. The V4 hypervariable region of the 16S rRNA gene was amplified and sequenced using Illumina MiSeq platform to characterize the microbial community structure and diversity. This research aims to validate the protective effects of Rothia mucilaginosa against constipation identified through Mendelian randomization analysis and understand how this intervention affects gut microbiome composition. The sequencing data will contribute to our understanding of microbiota-mediated therapeutic mechanisms and support the development of Rothia mucilaginosa as a potential probiotic candidate for constipation treatment.Additionally, we performed complete genome sequencing of Rothia mucilaginosa strain BNCC363031 (BeNa Culture Collection) used for the intervention. The genome was sequenced using PacBio Revio platform with HiFi (CCS) mode, generating high-accuracy long reads (>99.9% accuracy). De novo assembly using Hifiasm produced a single circular chromosome of 2,306,619 bp with 59.51% GC content. The complete genome provides genetic basis for understanding the probiotic mechanisms of this strain.
