Periplasmic stress contributes to a tradeoff between protein secretion and cell growth in E. Coli Nissle

To investigate changes in E. coli Nissle 1917 gene expression when engineered to secrete a large amount of protein via the curli (Type VIII) secretion system. To investigate variants of protein secretion and conduct gene expression profiling analysis to determine genes that are upregulated due to cellular burden from protein secretion. The different variants derived include: no protein expression or secretion, localized protein expression in the cytoplasm, and protein expression with secretion to the extracellular space (i.e., secretion through the inner and outer membrane).
