This study aims to investigate the impact of granulosa cell absence on the nuclear and cytoplasmic maturation of oocytes. Using the Smart-seq2 technique, single-cell RNA sequencing was performed on in vivo matured (IVO), cumulus-oocyte complex (COC), and denuded oocytes (DO) to compare the transcriptomic characteristics of oocytes under different culture conditions. By aligning and quantifying the data against the mm10 reference genome, this research obtained expression data of key genes and signaling pathways related to oocyte maturation and development. These findings provide insights into how defects in cytoplasmic maturation may potentially affect subsequent fertilization and embryonic development.
