The epitopic residue was determined to be critical for binding of mAb 20-8-4 to H-Kb because it did not bind to Kb with mutation K89A. The mAb does not bind to H-2Kbm3, which contains substitutions D77S and K89A. Mutation D77S did not affect binding. H-2Kb specific mAbs EH-144, B8-24-3, K9-136, and Y-25 demonstrated similar specificity.
