Renal Cell Carcinoma (RCC) associated with Xp11.2 translocation (TFE3-RCC) has been recently defined as a distinct subset of RCC. The Xp11 translocations involve the TFE3 transcription factor and produce chimeric TFE3 proteins retaining the basic helix-loop-helix leucine zipper structure for dimerization. To facilitate the development of molecular-based diagnostic tools and targeted therapies for TFE3-RCC, we generated a translocation RCC mouse model and performed DNA microarray analysis. A Rosa26-LSL-PRCC-TFE3 knockin (KI) mouse (PRCC-TFE3) was generated and crossed with KSP-Cre+ mouse. Kidneys were dissected out from 4 month old PRCC-TFE3; KSP-Cre(+) mice (n=4) , 4 month old PRCC-TFE3; KSP-Cre(-) mice (n=4), 7 month old PRCC-TFE3; KSP-Cre(+) mice (n=4) and 7 month old PRCC-TFE3; KSP-Cre(-) mice (n=3). Total RNA was isolated from flash frozen mouse kidneys using TRIzol reagent. cDNA preparation and hybridization of the probe arrays were performed according to the manufacturer's instructions (Affymetrix). Affymetrix GeneChip Mouse Gene 2.0 ST Arrays were applied. Partek Genomics Suite 6.6 was used for RMA-based data normalization and the subsequent data analysis, including PCA and ANOVA.
