Obesity is associated with chronic inflammation. H3K4me3 is generally described as a transcription activating histone modification and implicated in trained innate immunity and gene signature during lineage commitment of developing cels. We studied the effect of diet-induced obesity on H3K4me3 marks in hematopoietic stem progenitor cells. Using high fat diet-induced obesity in B6 mice as a model of pre-diabetic obesity, we studied H3K4me3 in LSK (Lin-Sca1+cKit+) and GMP (granulocyte macrophage progenitor: FcRg+CD34+Lin-Sca1-cKit+) hematopoietic stem progenitor cell (HSPC) populations in the bone marrow, compared with sex and age matched low fat diet-fed control mice. Downstream analysis was performed using Pluto Bio software (https://pluto.bio).
