Differential expression profile of liver tissue in rats with hepatic fibrosis

Liver fibrosis is a common pathological complication of end-stage liver disease, which is usually associated with chronic liver inflammation and injury. Liver fibrosis was induced by bile duct ligation (BDL) in rats. The differentially expressed genes in liver tissue of BDL rats were identified by microarray technique. In this study, the rat model of hepatic fibrosis was induced by bile duct ligation (BDL). Compared with the control group, the daily growth rate of the BDL group was significantly lower, whereas the ratio of liver wet weight to body mass of BDL group was significantly higher, respectively. The serum levels of ALT, AST, TBA and TBIL in the BDL group were significantly higher compared with those of the control group. The levels of deposition of collagen Ⅰ was assessed using Sirius red staining and the number of collagen fibers were assessed using Masson trichrome staining. The results showed that the rat model of hepatic fibrosis was successfully constructed. Then 3 liver fibrosis model rats and 3 normal rats were selected for microarray analysis,and differential gene expression in liver tissue of hepatic fibrosis was compared with that in normal liver tissue.
