Cutting propagation of peach trees is relatively complex, and adequate chilling improves survival rates, though mechanisms are not fully understood. Rooting phenotypic analysis revealed that the rooting rate of GF677 hardwood cuttings significantly increased after 30 days of sand storage, indicating it as the optimal for cutting. To further identify the genes associated with the chilling requirement for adventitious root (AR) formation, this study employed RNA-seq for comparative transcriptome analysis of cuttings treated with different durations of sand storage (0, 10, 20, 30 days). This study improved understanding of AR formation in peach cuttings under different chilling requirements and laid a theoretical foundation for technique optimization.
