Embryonic stem cell is a classical type of pluripotent cells that established from the inner cell mass of early embryos or blastocysts under appropriate culture conditions in vitro. ESCs have the ability to self renew indefinitely and the potential to differentiate into all cell types of the body. mESCs can share transcriptional and proteomic features of 2C embryos and display strikingly different patterns of pluripotency markers from the majority of ES cells in culture. Many critical regulation mechanisms can keep mESCs maintain only about 5 percent of cells transiently expressing Zscan4 or ERV and 95 percent of normally expressing pluripotent transcription factors such as Oct4 and Nanog. In recent years, significant progress has been made in understanding the molecular mechanisms underlying the regulation of two-cell gene expression in mESCs by epigenetic regulation.
