We analyzed expression changes between JAK2V617F positive bone marrow cells and JAK2V617F negative cells We further analyzed how TNF receptor (receptor 1 or receptor 2) block influenced these changes in JAKV617F positive relative to JAK2V617F negative Bone marrow was harvested from Balb/c mice with established JAK2V617F (GFP tagged) MPN. Cells were treated for 16 hours with our without TNFR1 or TNFR1 blocking antibodies (BAs). Cells were then sorted for Lin-Kit+ cells and subdivided into GFP+ and GFP- fractions and subject to RNA isolation and microarray analysis
