To identify RNA binding proteins critical for male gametogenesis, we performed RNA-Seq to search the RBPs specifically expressed in male gametocytes in the rodent malaria parasite P. yoelii. A double fluorescence reporters strain DFsc7, in which GFP and mCherry are specifically expressed in the male and female gametocytes respectively, was used for gametocyte induction in mice. Male and female gametocytes from DFsc7 were purified by fluorescence-activated cell sorting (FACS) and subjected to RNA-seq to generate gender-specific transcriptomes.
