Mice infected with P. berghei were treated with sub-curative doses of Primaquine (PQ) over 8 passages. Unlike other similar drug pressure assays performed at Imperial (e.g Lumefantrine), cloned parasites with confirmed resistance to PQ also showed an inability to transmit to mosquitoes. It is hypothesized that PQ resistance mode-of-action is linked to the production of non-viable gametocytes (similar to atovaquone which was recently reported). Consequently, evolution of PQ resistance is hypothesized to be self-limiting as it cannot escape the mammalian host and complete life cycle in the mosquito.
