We aimed to investigate the role of XPO1 in this rare leukemia subtype. RNA-seq was performed in DEK::NUP214 expressing cell line in control and eltanexor-treated samples, an inhibitor of XPO1, to elucidate the genes and pathways involved in DEK::NUP214 AML To investigate the role of XPO1 in DEK::NUP214 leukemia, cell line and PDX model expressing the fusion gene were treated with the XPO1 inhibitor eltanexor. Gene expression profile was investigated after treatment. .The human FKH-1 cell line was cultured in vitro in RPMI supplemented with 20% FCS. Cells were treated with 400nM eltanexor or vehicle (DMSO) for 24h. Total RNA was isolated using the Rneasy kit from Qiagen, followed by DNAse treatment before sending for sequencing. Human AML cells isolated from bone marrow of DEK::NUP214 mice (PDX1) and sorted for expression of hCD45+ cells after 11 days of treatment with vehicle or eltanexor 12.5 mg/kg
