In situ HiC of primary mouse B cell progenitor populations after activation with LPS

Naive resting B-cells were purified from the bone marrow of C57BL/6 mice using flow cytometry sorting. Naive B cells were activated with LPS. Activated but undivided cells were harvested 3 hours, 10 hours or 33 hours after the LPS treatment. The Fluorescent Ubiquitination-based Cell Cycle Indicator (FUCCI) system was also used to isolate imminently dividing B cells specifically in the G1 stage of the cell cycle prior to the S phase. Two biological replicates were prepared at each activation time. In situ Hi-C libraries were sequenced on an Illumina NextSeq 500 to produce paired-end 81bp reads.
