The goal of this study was to identify genes whose loss induced enzalutamide sensitivity in castration-resistant prostate cancer cells (C4-2B). One third of a commercially available bar-coded lentiviral shRNA library (DECIPHER from Cellecta, module 1) was transduced into C4-2B cells, targeting approximately 5,000 genes. Cells were transduced with lentiviral pooled shRNA library and then each line will be split into three groups. Cells in the first group were collected and represent the initial population of shRNA quantity . The cells from the second group served as an enzalutamide-negative control group (cell culture in the enzalutamide vehicle control [DMSO] for 6 days), and cells from the third group were cultured in the presence of enzalutamide for 6 days . Genomic DNA was isolated and shRNA barcodes were quantified by sequencing.
