Genetically modified pigs are commonly created by using somatic cell nuclear transfer (NT). Treatment of reconstructed zygotes with histone deacetylase inhibitors (HDACi) immediately after NT improves cloning efficiency. The objective of this experiment was to compare the effectiveness of Suberoylanilide Hydroxamic Acid (SAHA) and its hydrophobic derivative 4-iodo-SAHA (ISAHA) to Scriptaid. SAHA and ISAHA can successfully be used to create healthy piglets from NT. Transcriptome analysis of blastocyst stage embryos identified changes in gene expression IVV, IVF and NT embryos either treated with SAHA, ISAHA, Scriptaid or untreated.
