High-throughput determination of let-7a and miR-21 loaded RISC cleavage kinetics

Argonaute proteins loaded with small interfering RNAs (siRNAs) or microRNAs (miRNA) form the RNA-Induced Silencing Complex (RISC), which represses target RNA expression. We applied high-throughput methods to measure single-turnover cleavage rates of RISC in vitro and to measure knockdown of thousands of RISC targets in an engineered cellular system.
