资源简介:
Nanopore current recordings for the Nature communications publication titled "Digital Immunoassay for Biomarker Concentration Quantification using Solid-State Nanopores" by He & Tessier et al., in which we present a digital immunoassay scheme capable of reliably quantifying the concentration of a target protein in complex biofluids that overcomes specificity, sensitivity, and consistency challenges associated with the use of solid-state nanopores for protein sensing. This is achieved by employing easily-identifiable DNA nanostructures as proxies for the presence (“1”) or absence (“0”) of the target protein captured via a magnetic bead-based sandwich immunoassay. As a proof-of-concept, we demonstrate quantification of the concentration of thyroid-stimulating hormone from human serum samples down to the high femtomolar range. The raw nanopore current recordings collected for each biomarker concentration are provided here. The ionic current recordings were performed in MATLAB 2013a (32-bit) using the VC100 current amplifier (Chimera Instruments) with sampling frequency of 4.17 MHz and a bandwidth of 1 MHz, and were subsequently software low-pass Bessel filtered as needed. Further information is available in the README.txt file and the associated publication.
