Project goal. Whole genome and transcriptome analyses of the black scraper (Thamnaconus modestus). Genome sequencing of individual marine animals using the PacBio-Sequel and the HiSeq Paired-end methods. Transcriptome sequencing using the HiSeq Paired-end and the IsoSeq-Sequel sequencing methods, and DEG analysis using templates prepared from individual marine animals at varying time points after artificial infection or stimulation . Acquisition and analysis of useful genes. Selection of 15 or more useful genes from each marine animal. Expression analysis of 8 or more genes from the selected useful gene pool. Production of three or more of the selected useful gene products by recombinant expression systems or peptide synthesis and ligation to identify their functions ''Project contents. Production of whole-genome data from the black scraper. Genomic DNA extraction from a black scraper population. Approximately 0.5 Gb of data production using the PacBio-Sequel (>20 kb) sequencing. Approximately 25.9 Gb of data production using the HiSeq Paired-end (2 x 101 bp) sequencing . Production of the black scraper transcriptome data. 34 samples isolated and prepared at different time periods after artificial infection or stimulation for differential gene expression analysis. Production of 7.7 Gb of data per sample using the HiSeq Paired-end (2 x 101 bp) sequencing. Approximately 48.6 Mb of data production using the IsoSeq-Sequel sequencing ''Expected efficacy and applications. Providing technology to aid academic and industrial use of major aquaculture and aquatic biological resources through the completion of aquatic standard dielectric and establishment of a specialized genomic data platform for aquatic organisms. Accelerating research on aquacultured species by establishing economical methods to identify useful genes and gene markers.
