Effect of plant genotype and production site on microbiota of mature seeds and seed exudates

We studied the effects of plant genotypes (8 French commercial bean cultivars) and production zone: Gers in South-west of France and Maine et Loire in the Loire Valley; on the microbiota (bacteria and fungi) on mature seeds and seed exudates. The seed exudates were collected after 2h, 8h, 16h and 24h of seed imbibition, and we also analyzed a pool of all the time points. The microbiota was studied using amplicon sequencing. For the mature seeds, we used 16S rRNA gene (V4 region) and gyrB gene to characterize the bacterial community. We used ITS1 region for the fungal community. For the exudates, we used the gyrB and ITS1 markers.
