MTA1 promotes prostate cancer progression by upregulating androgen receptor splice variant-7 (AR-V7)

Androgen receptor (AR) signaling is a critical pathway in prostate cancer (PCa) cells, and andro-gen receptor splice variant-7 (AR-V7) production is crucial in PCa progression, metastasis, and drug resistance. We confirmed the effects of metastasis-associated protein 1 (MTA1) on PCa cell proliferation and migration, and used the first transcriptome sequencing of 22RV1 MTA1 knock-down cells to identify the associations of MTA1 with AR and AR-V7, which were validated at the protein and RNA levels. Furthermore, we assessed the effect of MTA1 expression on cell sensi-tivity to the AR inhibitor enzalutamide. We found that MTA1 was highly expressed in PCa and positively correlated with its clinical stage. Furthermore, MTA1 was shown to induce PCa pro-gression by upregulating AR and AR-V7 while also promoting AR splicing to AR-V7, which led to cell resistance to enzalutamide. Overall, this study provides novel insights into MTA1 as a po-tential therapeutic target for PCa.
