This dataset contains the source data for the manuscript: "Sodium dodecyl sulfate triggers rapid human skin barrier disruption through oxidative stress." The study investigates the rapid impact of sodium dodecyl sulfate (SDS) on human skin barrier integrity using an ex vivo human skin model (NativeSkin®). This dataset provides the key multi-omics data generated, including proximity extension-based proteomics (Olink®) and processed RNA sequencing (RNA-seq) data, from skin explants exposed to different concentrations of SDS and antioxidant treatments (N-acetylcysteine [NAC] and nicotinamide [NAM]). The data files correspond to short-term (5-minute) and prolonged (6-hour) SDS exposures, as well as the antioxidant recovery experiments. These data were used to identify molecular signatures of barrier disruption, including the induction of oxidative stress, endoplasmic reticulum (ER) stress, and epidermal differentiation pathways. Data Description The data is organized by experimental setup and data type: 1. Proteomics (Olink) - 5-minute SDS Exposure (Figure 1 Data) olink_NPX_5min_exposure.xlsx: Olink NPX data for ex vivo skin tissue lysates 24 hours after a 5-minute SDS exposure (0, 1, and 5 mg/mL). olink_metadata_5min_exposure.csv: Metadata describing the samples used in the 5-minute Olink experiment. 2. Proteomics (Olink) - 6-hour SDS Exposure (Figure 2 Data) olink_NPX_tissue_6h_exposure.xlsx: Olink NPX data for ex vivo skin tissue lysates 48 hours after a 6-hour SDS exposure (0, 0.04, 0.2, 1, and 5 mg/mL). olink_NPX_supernatant_6h_exposure_inflammation.xlsx: Olink NPX data (Inflammation panel) for culture supernatants 48 hours after a 6-hour SDS exposure. olink_NPX_supernatant_6h_exposure_immune_response.xlsx: Olink NPX data (Immune Response panel) for culture supernatants 48 hours after a 6-hour SDS exposure. olink_metadata_tissue_supernatant_6h_exposure.xlsx: Metadata describing the samples for the 6-hour Olink experiments (both tissue and supernatant). 3. Transcriptomics (RNA-seq) - 6-hour SDS Exposure (Figure 3 Data) rna_raw_counts_6h_exposure.csv: Raw gene-level counts for ex vivo skin 48 hours after a 6-hour SDS exposure (PBS, 1, and 5 mg/mL). rna_normalized_counts_6h_exposure.csv: Normalized counts (DESeq2) for the 6-hour exposure experiment. rna_fpkm_values_6h_exposure.csv: FPKM (Fragments Per Kilobase of transcript per Million mapped reads) values for the 6-hour exposure experiment. rna_metadata_6h_exposure.csv: Metadata describing the samples for the 6-hour RNA-seq experiment. 4. Transcriptomics (RNA-seq) - Antioxidant Treatment (Figure 5 Data) rna_counts_antioxidant_treatment.csv: Raw gene-level counts for the antioxidant recovery experiment (SDS, SDS+NAC, SDS+NAM). rna_normalized_counts_antioxidant_treatment.csv: Normalized counts (DESeq2) for the antioxidant experiment. rna_fpkm_combatseq_antioxidant_treatment.csv: ComBat-seq batch-corrected FPKM values for the antioxidant experiment. rna_metadata_antioxidant_treatment.csv: Metadata describing the samples for the antioxidant RNA-seq experiment.
