Intestinal organoids are three-dimensional (3D) polarized structures composed of different types of cells capable of self-organization and self-renewal, resembling their organ of origin in architecture and function. Organoids represent a significant development due to their organised multicellular structure that more closely represents in vivo tissues than any other cell culture technology previously developed. To facilitate study, refine and validation of the different organoid cultures systems need to be performed. Polarisation of our gut enteroids was inverted by EDTA treatment and media change. RNA extraction and sequencing were carried out to determine potential changes in gene expression before and after polarity inversion. We establish that polarity change did not affect cellular integrity in bovine and ovine basal and apical out organoid cultures. Therefore, apical-out organoids appear to have many advantages to be used as new in vitro models due to their direct accessibility of the apical surface.
