Testing the applicability of tSNE to 27-parameter single-cell flow cytometry data. Conclusion: Background fluorescence and spreading error impact the visualisation of multidimensional single-cell data by tSNE substantially, specifically resulting in the generation of virtual “islands” that do not reflect real heterogeneity at the phenotypic or biological level.
