mRNA profiling on GM-CSF knockout CART19 compared to wildtype CART19 and Ctrl gRNA CART19 produced from normal donors

RNA sequencing was performed on three biological replicates of CART19 (GM-CSF wildtype), GM-CSF knockout CART19, and Ctrl gRNA CART19 produced from normal donors. T cells from normal donors were transduced with a replication-incompetent lentiviral vector expressing a second-generation chimeric antigen receptor (CAR) consisting of an anti-CD19 single chain variable fragment (FMC63) fused to 4-1BB and CD3ζ intracellular domains as previously described. GM-CSF knockout CART19 were also transduced with CSF2 CRISPR Guide RNA in a pLentiCRISPR v2 backbone as previously described. Ctrl gRNA CART19 were also transduced with non-targeting (Ctrl) guide RNA in a pLentiCRISPR v2 backbone as previously described. RNA was isolated on Day 8 of production.
