In this study, the transcriptomes of roots of two-day-old cft mutant and wild type A17 seedlings were generated. Briefly, RNA was isolated from roots of cft mutant and wild type A17 seedlings that had been growing vertically on square Petri dishes (120 x 120 mm) with half strength of Murashige & Skoog (1/2 MS) basal salt mixture in a growth chamber at 22 °C with a 16-h-light/8-h-dark regime (100 µmol m-2. s-1) for two days. The RNA was then treated with DNase to remove potential DNA contamination. The RNA-Seq libraries were constructed using TruSeq Stranded mRNA Sample Preparation kits (Illumina) following the manufacturer's protocols. The RNA-Seq libraries were sequenced in an Illumina Hi-Seq2000 sequencer.
