This study was designed to compare the global gene expression change induced by the circulating, prodomain bound forms of BMP9 and BMP10 (pro-BMP9 and pro-BMP10) in human pulmonary arterial endothelial cells (PAECs). This is different from many previous studies which used the growth factor domain of BMP9 and/or BMP10. Four independent PAEC lines were purchased from Lonza. Each line was grown up and seeded separately for the treatment. BMP samples were made up on the day of the treatment, and cells were treated at 0.3 ng/ml of final growth factor domain concentration. Treatment was carried out for 1.5 hours at 37 C to detect the genes directly regulated by pro-BMP9 and pro-BMP10.
