To characterize our compromised reconstructed epidermis at the transcriptomic level, we have employed whole genome microarray expression profiling as a discovery platform to compare genes modification with those known in AD skins. To this purpose, normal human keratinocytes (NHK) were used to generate RE, treated or not with an inflammatory cocktail (Poly I:C, TNF-α, IL-4 and IL-13). Inflammation and cellular differentiation pathways were analysed and then confirmed by Q-PCR experiments. Inflammation induced gene expression in reconstructed epidermis was measured 2 days after exposure to inflammatory cocktail or normal medium. Four independent experiments were performed for each condition (inflamed or normal) using different donors for each experiment. Contrinuted by: R&D department, SILAB, France
