CSRP2 is an actin-bundling protein essential for assembly of invadopodia. We observed that it is also present in the nucleus and its knock-down results in decreased expression of MMPs important in the invasive or metastatic process. Therefore, we sought to examine the transcriptomic impact of loss of CSRP2 in human breast cancer cells. 150 000 Hs578T cells were plated in each well of a six-well plate. The following day, Lipofectamine 2000 was used to transfect the cells with non-targeting or CSRP2 siRNA. 24 hours later, 100,000 cells from each well were transferred to a 24-well plate. 48 hours later, RNA was extracted from the cells and analyzed on Affymetrix HuGene 2.0 arrays.
