We investigated transcriptional responses following in vitro stimulation of peripheral blood naive CD4+ T cells and naive B cells obtained from a patient with inherited c-Rel deficiency. Samples from unrelated healthy donors and a patient with CD40 deficiency were analyzed for comparison. Overall design: RNA seq was performed on (i) isolated naive CD4+ T cells from 4 healthy donors and a patient with AR c-Rel deficiency, 2 hours after activation with PMA, or with different combinations of coated beads with anti-CD3, anti-CD2 or anti-CD28 Mabs; (ii) isolated naive B cells from 4 healthy donors, a patient with AR CD40 deficiency and a patient with AR c-Rel deficiency, 2 hours after activation with PMA, CD40L, or F(ab')2 goat anti-IgM.
