MTT Cell Proliferation Assay kit from ATCC (Cat. No. 30-1010K). The assay is based upon measuring the reduction of tetrazolium salts to examine changes in cell proliferation. The yellow tetrazolium MTT (3-(4, 5dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide) is reduced by metabolically active cells, in part by the action of dehydrogenase enzymes, to generate reducing equivalents such as NADH and NADPH. The resulting intracellular purple formazan crystals that form in metabolically active cells can be solubilized using a detergent solution and quantified by spectrophotometric means.
