The aim is to know transcriptional profile change in RpoB_A621E mutants versus its parent strains. This mutation though to be responsible for the reduced susceptibility to both vancomycin and daptomycin in S.arueus. A total of four comparisons were performed by comparing bacterial strains of the Daptomycin- and vancomycin-susceptible wild type strain N315?IP (native RpoB) vs the following four strains: 1) Daptomycin-selected dual daptomycin- and vancomycin-resistant strain 10*3d1 (harboring 4 gene mutations including rpoB_A621E mutation); 2) Gene-replacement mutant N315?IP-rpoB_621E; 3) Gene-replacement mutant 10*3d1-SA1862_14A-rpoB_621A and 4) 10*3d1-SA1862_14A-rpoB_621E (See Ref.2) RNA extraction and microarray hybridization was performed as described in Ref.1.
