To explore the role of epidermal keratinocyte STAT3 in the pathogenesis of atopic dermatitis (AD), we developed tamoxifen-inducible K14-CreERT-Stat3flox/flox mice. After intraperitoneal injection of tamoxifen for 5 consecutive days, these mice became epidermal keratinocyte-specific STAT3 deficiency. After tamoxifen induction, DNCB was topically applied to the ears of the mice to simulate AD-like skin inflammation. RNA sequencing was performed on the total epidermal RNA from mouse ear skin after being treated with DNCB or vehicle on 21 days.
