In this study, we aimed to investigate the differentiation dynamics of megakaryocyte development, utilizing human umbilical cord blood (UCB) cells Overall design: We cultured human umbilical cord blood (UCB) cells to generate megakaryocytes by first isolating mononuclear cells from UCB using density gradient centrifugation, then plating the cells in a serum-free medium supplemented with thrombopoietin (TPO) to promote megakaryocyte differentiation, maintaining the cultures at 37°C in a humidified atmosphere with5% CO2, and periodically assessing cell morphology and surface markers using flow cytometry to confirm the generation of megakaryocytes
