Whole-genome RNAsequencing of intestinal organoids and tumoroids

In order to decipher the molecular responses of normal epithelial cells to the tumor secretome, we obtained the gene expression profiles of wildtype organoids exposed to either WT or tumor conditioned medium, along with the transcriptional signature of the tumoroids from which the corresponding tumor conditioned medium was derived. Overall design: Three replicates of wildtype organoids (from 3 different mice) exposed for 24h to conditioned medium (wildtype or tumor organoids) and three replicates of tumoroids (from 3 different mice) grown for 24h were used for the analysis. A total of 9 samples was analyzed (3 per condition).
