Single-cell RNA sequencing of human salivary glands and organoids

We performed single-cell RNA sequencing (scRNA-seq) on human salivary gland tissues and derived organoids to investigate their cellular composition, differentiation states, and regenerative potential. Four samples were dissociated into single-cell suspensions and processed using the 10X Genomics Chromium platform. Overall design: Human salivary gland tissues and organoids at defined culture timepoints were enzymatically dissociated, viability-checked by AO/PI staining, and subjected to scRNA-seq using the 10X Genomics Chromium 3' v3 system.
