To investigate key differentially expressed genes (DEGs) across various stages of (hepatic ischemia-reperfusion injury) HIRI, our study employed the mouse HIRI model and performed whole-transcriptome sequencing with liver tissues sampled at sequential HIRI stages. All experimental and control mice were 8-to10- week-old male C57BL/6J mice (specific pathogen-free) with body weights ranging from 20 to 25g. This study included 32 mouse liver samples, which were divided into 4 groups based on different ischemia times, with 8 samples in each group. The control group (8 mice) received anesthesia and laparotomy only. Eight mice underwent 1 hour of ischemia and were sacrificed without restoring the blood supply (0 h group). This phase represents the period before reperfusion begins, during which liver injury is relatively mild and is solely caused by ischemia without involving reperfusion factors. Eight mice underwent 1 hour of hepatic inflow occlusion followed by 6 hours of reperfusion (6 h group), a time point corresponding to peak liver injury severity and inflammatory response. The final 8 mice underwent 1 hour of portal triad occlusion, after which the blood supply was restored and maintained for 24 hours (24 h group). This group represents the recovery phase of HIRI, with significantly reduced tissue damage and attenuated inflammatory response compared to the 6 h group.
