To assess the function of FoxO4 in processes of TH1 differentiation, we have performed a whole genome microarray assay to discover the significant changed genes in normal and FoxO4 overexpressing TH1 cells. Isolated naive CD4+CD25−CD62LhiCD44lo T cells from 8-week-old C57BL/6 mice were activated with anti-CD3 and anti-CD28. 20 hours later, cells were infected with RV or RV-FoxO4, and cultured in TH1 polarizing condition for 72 hours. Next, total RNA was extracted from sorted GFP positive cells, and Agilent-028005 SurePrint G3 Mouse GE 8x60K Microarray were used to probe global gene expression changes.
