The goal of this study was to see if isolating CD8 T cells from PBMCs with a DNA aptamer versus an antibody would have 1) any immediate transcriptional affect on freshly isolated T cells and 2) any downstream transcriptional affect on a manufactured CD19 CAR T cell product. There are a total of 18 samples, divided into three groups of 6 by day and cell type (D0, S1R1D14 mocks, S1R1D14 CD19 CAR). Within each group of 6, there are 3 biological replicates (i.e. separate donors) for both aptamer- and antibody-isolated cells. Furthermore, the 3 separate donors are the same between aptamer- and antibody-isolated cells and across the groups, enabling pair-wise comparisons. The antibody-isolated samples were thus considered the control/reference samples to which the aptamer-isolated samples were compared (donor-by-donor within each group) to.
