Vascular endothelial injury caused by post-hemorrhagic shock mesenteric lymph (PHSML) return is an important cause of refractory hemorrhagic shock, but the underlying mechanisms need to be further studied. The present study investigated the molecular mechanism of PHSML on the injury of human umbilical vein endothelial cells (HUVECs) based on transcriptome analysis. PHSML was collected from male rats underwent hemorrhagic shock and following resuscitation. The normal mesenteric lymph (NML) was harvested from the sham rats. HUVECs were incubated with the culture medium containing 10% phosphate buffered saline (Control), NML and PHSML respectively. After incubation for three hours, the cells were harvested for RNA sequencing (RNA-seq).
