Two historical samples labeled as “Variola vera” have been deposited in the National Museum of Prague, Czech Republic, for more many years. Small pieces of tissue were taken from both specimens for DNA/RNA isolation, tandem mass spectrometry analysis of protein composition, histological, electron microscopic and chemical composition analyses. DNA from the samples was analyzed by whole genome sequencing. The genome provide further insight into the strain of variola virus that might have circulated in Europe at that time.
