Dendrobium is one of the largest genera in Orchidaceae, and is also an important ornamental orchid genus, but its high-quality genome has not been previously published. Here, we report a chromosome-level genome of D. thyrsiflorum. We reconstruct the chromosome evolution of Dendrobium orchids and the gene family expansion and contraction of the D. thyrsiflorum genome, which is the candidates involved in the pathways of their secondary component biosynthesis and environment adaption. Then, we also identify the gene involved in their horticultural value like the MADS-box involved in floral organ development, TPS gene family for their secondary component and environmental adaptation, and the genes involved in the pathways of color formation in D. thyrsiflorum. Our results provide valuable genomic resources for the improvement of orchids and other ornamental plants.
